遊戲中不只是收錄吸血鬼獵人一方的正派角色，同時也包括吸血鬼之王德古拉等反派角色，目前已知的登場角色包括初代作主角吸血鬼獵人西蒙，具備神秘聖獸之力的少女瑪莉亞，憂鬱的半吸血鬼貴公子阿爾卡特，以及一切災厄根源的吸血鬼之王德古拉。
遊戲最大的特色在於與以往系列作大不相同的玩法，從單人打怪變成雙人對戰，是系列作首度嘗試。對戰時基本上是以揮動Wii遙控器的輕攻擊為主力，並搭配重攻擊來施展連續技，副武器可用來進行牽制，並伺機累必殺技計量表以強力超必殺技一決勝負。 |
